The project focuses on the development of multifunctional nanorobots for targeted, non-viral, and minimally toxic gene engineering of immune cells. The nanorobots will combine advanced nanomaterials, targeted surface functionalization, and magnetic, enzymatic, or light-driven propulsion to efficiently deliver nucleic acids and CRISPR-based gene-editing components directly into T and NK cells. This approach aims to overcome major limitations of conventional viral transduction and electroporation, including immunogenicity, off-target integration, and high cellular toxicity.
The project will use the developed nanorobotic platform to generate advanced CAR-expressing T and NK cells with improved cytotoxicity, persistence, and reduced immunosuppression. By improving the safety, precision, and scalability of immune-cell engineering, the technology could contribute to the development of next-generation cancer immunotherapies. Beyond oncology, the platform may also have future applications in autoimmune diseases, infectious diseases, and transplantation medicine.
